CFG Gene Targeting Services

The CFG provides technologies for in vivo gene function analysis and generation of animal models produced via gene targeting in Embryonic Stem (ES) cells with subsequent blastocyst injections. This basic service is divided into two stages. Electroporation of ES cells and Blastocyst Injections. Both or either one of these services are offered at CFG.

Electroporation of Embryonic Stem Cells

This includes electroporation of DNA into embryonic stem cells, positive/negative selection, collection of 200 embryonic stem cell clones and screening. Individual clones with duplicates will be isolated for DNA analysis. Targeting will be either in ES cells derived from129Sv genetic background or W4 ES cells, derived from 129SvEv.

To Request Services for Electroporation

  1. For first time users, complete and submit a Customer Information Form that you can download directly in either word or acrobat formats.
  2. Complete and submit an ES Cell Form that you can download directly in either word or acrobat formats.
  3. Complete and sumit the ES cell clone screening form that you can download directly in either word or acrobat formats.
  4. Arrange to send DNA construct(s) to the CFG. Please refer to Instructions for DNA Cleaning and Shipping. [word] or [acrobat]

We recommend isolating the genomic DNA target from the same genetic background as the ES cells to be used. We offer two different genomic libraries to screen; a cosmid library derived from 129Sv, and a lambda library derived from129 SvEv genetic background. Blastocyst Injections

ES cell clones that have been karyotyped will be injected into C57Bl6 blastocysts. Generation of at least 2 chimaeric mice is guaranteed. Mice are kept in SPF conditions within barrier facilities maintained by Taconic Farms, Inc.

  3. Complete and submit the Mouse Screening Form that you can download directly in either word or acrobat formats.
  4. Complete and submit the Mouse Shipping Form that you can download directly in either Word or Adobe formats. Shipping will be arranged through Taconic Farms, Inc.

The investigator may send their own targeted ES cell clones for injection. However, we cannot guarantee germline transmission for these clones.
